EXTREMA is a high performance liquid chromatograph (HPLC) that brings together the best of JASCO’s technology. You can build the optimal system for your purpose from a variety of units, including UHPLC, RHPLC (Rapid Separation HPLC), semi-preparative pumps, and high-sensitivity detectors. The chromatography data system, ChromNAV, renowned for its operability and processing functions, now offers enhanced functionality and user-friendliness with its upgrade to Ver. 2.0. This version includes various improvements, making it more efficient and intuitive for users.
System lineup
There are three types in the lineup: HPLC/RHPLC, UHPLC, and Prep-HPLC. Many units are available, and you can build the optimal system by combining units suited to your purpose.
UHPLC
Ultra High Performance Liquid Chromatography (UHPLC) uses columns packed with ultrafine particles of 2µm or less, enabling separation at high pressure (up to 130 MPa). This method allows for high-speed separation while maintaining high separation efficiency. Compared to conventional HPLC, UHPLC significantly shortens analysis time, reducing it to just 1/5 to 1/10 of the usual duration. Additionally, it minimizes the amount of mobile phase solvent used, cutting it down to nearly 1/10, thus enhancing both efficiency and sustainability.
HPLC/RHPLC
Rapid Separation HPLC (RHPLC) provides a high-speed analysis method that spans the range from conventional HPLC to some UHPLC applications. This analytical system utilizes fully porous packed columns with particle sizes of 2 to 3 µm and core-shell columns. Moreover, it supports measurements in the medium pressure range (30 to 70 MPa), positioning itself as the future standard analytical method.
Prep-HPLC
Preparative High Performance Liquid Chromatography (Prep-HPLC) focuses on isolating and purifying target compounds with high purity from synthetic products and natural substances. EXTREMA, equipped with a semi-preparative pump, enables its use as a semi-preparative high performance liquid chromatograph (Prep-HPLC). Additionally, it facilitates micro- and ultra-micro-preparative separations when integrated with an RHPLC or UHPLC system.

Hardware
Pump
The pump includes an expansion bay, enabling the installation of various units inside the housing for compact and functional system upgrades. With a total of 10 models available, it caters to HPLC/RHPLC, UHPLC, and preparative use.
Pumps for HPLC/RHPLC, UHPLC
The PU-4100 and 4200 models employ the Advanced Slow Suction Quick Delivery (SSQD) method, ensuring stable delivery across a range of flow rates. This method achieves a delivery performance with a flow rate precision of 0.05% (RSD value).
The PU-4285 pump offers a pressure resistance of 130 MPa, making it suitable for a wide range of applications from conventional HPLC to UHPLC. The PU-4180/4185 pumps, with a pressure resistance of 70 MPa, are compatible with both conventional HPLC and RHPLC. Additionally, the PU-4185/4285 models provide a binary pump option for high-pressure gradients, enhancing versatility and performance.
Semi-preparative pump
The PU-4086 supports a maximum flow rate of 20mL/min, and the PU-4087 supports a maximum flow rate of 50mL/min. The motor control technology developed for semi-preparative pumps and the high torque motor have significantly improved the maximum operating pressure at high flow rates. This allows for high flow rates even for preparative columns using 5µm diameter packing materials.
Additionally, one can install a recycle unit for recycling and separation purposes. This recycle unit fits conveniently inside the pump’s expansion bay.
Highly expandable pump
The expansion bay in the pump can accommodate a degassing unit, low pressure gradient unit, solvent switching valve unit, mixing unit, manual injector unit, etc. In the case of a single pump, a second pump unit can be installed.
Software Features
System control using ChromNAV Ver.2
ChromNAV Ver.2 is an easy-to-use chromatography data system. It fully utilizes the performance of EXTREMA, from controlling the entire system to analyzing measurement results and managing data.
ChromNAV CFR Ver.2 with enhanced data integrity is also available.
Data acquisition and analysis using ChromNAV Lite
ChromNAV Lite is a PC integrator with excellent operability. It is specialized for recording and analysis functions, and has high cost performance and a simple and easy-to-understand user interface.
Measurement example
Analysis of fatty acids by rapid separation HPLC (RHPLC)
Rapid Separation HPLC (RHPLC) is a high-speed analysis method that covers the range from conventional HPLC to some UHPLC. Here, we used an RHPLC system to measure eight fatty acid components using conventional HPLC and UHPLC columns.
When fatty acids were measured using a refractive index detector, the analysis time was reduced by approximately half when using a UHPLC column with a 1.9 µm particle size (B) compared to when using an HPLC column with a 5 µm particle size (A).
Analysis of fat-soluble vitamins
Vitamins, one of the five major nutrients, are essential for keeping the body in good condition. Among them, fat-soluble vitamins are found in the living organisms of animals and plants and are involved in various physiological functions in metabolism. Here, we used a PDA detector to analyze eight fat-soluble vitamin components.
The chromatogram and contour lines of the fat-soluble vitamins are shown in Figure 2. The eight components were successfully separated within 25 minutes.
Analysis of ibuprofen using a circular dichroism detector
Ibuprofen is a type of nonsteroidal anti-inflammatory drug that is widely distributed in Japan both as a prescription drug and over-the-counter drug. Ibuprofen is an optically active substance that exists in both R- and S-forms. Here, we analyzed ibuprofen using a CD detector and an optical separation column.
Figure 3 shows the chromatogram of a mixed sample of caffeine and racemic ibuprofen. In the UV chromatogram, caffeine is also detected, but in the CD chromatogram, it can be seen that only ibuprofen, an optically active substance, is selectively detected. Comparing the spectra, it can be seen that only ibuprofen shows CD (Figure 4).
